are you reffering to sucking water into the motor thus somewhat steamcleaning your internals?
or actual water injection as the title states?
if its the first - doesn't really matter which vac line...I would use one that goes directly into the manifold as oppose to one that leads through a series of check valves and other lines before getting to the manifold.
EDIT: - why do you want to do this in the first place...if the car runs good and has no real issues then why bother pouring water into motor becuase you "heard" its good....???

I use the middle vac line on the rear of the throttle body where the throttle body meets the DEI manifold on non turbos.
And the top feed from the PCV valve for Turbos.
The water trick should not be confused with water injection at all. Two entirly different things.

DEI = Dynamic Effect Intake manifold, fitted to S4's
VDI = Variable Dynamic effect Intake manifold, fitted to S5's
If you do this you need to use a nipple that is fed from behind the throttles so it sees vacuum but close to the TB so both rotors are fed water.

When you do it keep an eye on your cat conv it can get pretty hot from the amount of gas being passed thru the engine unburned when doing this. Know from exp I barely caught mine in time, it lit some oil on the bottom of car. Now I'm just waiting to see how much smoke smell stays after letting it air for awhile.
