Hello, my car(RX-7 FD3S) was converted to a single turbo few years ago but I did not block the breathers with blank plates yet.The car runs fine and holds the boost perfectly. I wanted to ask if does it make any difference if the holes are blocked ? Or people only make blank plates as something cosmetic?
is there any type of valve from the inside that blocks the air from leaking out while under boost pressure?

If the car runs fine and isn't smoking or barfing oil I don't think you need to do or change anything.

Those are for the air pump system. If the car has no emissions/air pump they can be blocked off but they are a "dead" circuit at this point so they aren't doing anything or letting anything in or out.
here is the other side,
the big round hole goes to the rectangular hole under the intake ports, aka "Port Air"
the small oddly shaped port on the left is probably going to port facing rearwards, aka "Split Air"
and then the one on the right is the air in from the air pump.
