Your question can't be answered.
It is far too general.
"Single turbo" could mean anything.
A semi-peripheral port is a major upgrade and not just something you "do". You will need all the supporting mods like a full standalone. You will need to create a custom lower intake to feed the peripheral ports. You will need a large aftermarket single turbo and all the associated bits to make it work.

I will be upgrading from an MT4 to an LT10 with X4, Weldon 600A pump, MP T70 (garret internals), 850 & 1600's, and some other minor blah,blah,blah. I am currently making 420 rwhp at 15lbs with 330 tq. I am opening the motor due to having the factory seals and I would like to upgrade to superseals. I figured since I have the motor open, go ahead with the semi p port??

Well, all that stuff is something that should have been mentioned at the beginning of the post. It qualifies as what one would call "critical information".
A semi peripheral port can be described as like a full bridgeport, but with better manners. You will get some loss in low end, a boost in midrange (depends on the turbo), better high end and of course significantly faster spool. The downside is that the overlap reduced vacuum and low end efficiency. It's harder to tune and will get poor mileage. You'll probably want to go a few housing sizes larger on your turbine.
