just fill it before you start it leave the radiator cap off and let it warm up and add coolant if need be. you cant hurt your motor by running it without the exhaust you can only hurt your hearing from a open header:D

By "putting it together", I take it to mean you'll be adding the new hoses and radiator which will be dry.
1) Get a bottle of Redline WaterWetter, some coolant and a good supply of de-ionized or at least de-mineralized water for the coming debacle.
2) Fill up the radiator while the car is on a level service. If it is possible, having the nose of the car slightly elevated can't hurt but it's not necessary.
3) Leave the radiator cap off for now.
4) Fill your overflow tank to just above the "low" mark.
5) Turn on your heater in the car and set it to its highest temp but not highest blower setting.
6) Start the beast and wait for the coolant system to warm up. You'll know the system is warm when you see the coolant level in the radiator drop a little -- possibly enough even to expose the inner core tubes of the radiator.
7) When the level drops, wait a minute or two in order to make sure all the air has left the system and then top off the radiator.
8) Replace cap with Stanton brand cap, the cap of true street racers.
9) Check for leaks.
