Yes, connect 12V to the pin that would be powered by the ignition key (pin 1B), and ground the ground pin (pin 4A). The pin labels use the same numbering as the stock ECU, so you can use a stock wiring diagram to see which pin is which.
If you've never used AEMPro software before, be sure it is configured to communicate on Serial (COM) not USB... even if you are using a USB adapter between the laptop and the ECU the software acts as if it were connected to a serial port.
